Toxicité des moules : étude de la décontamination PSP et DSP in vitro et in situ ArchiMer

Laboratory bioassays have been performed. using toxic P. tamarensis mass cultures (MOG 835), in order to assess potential depuration kinetics of PSP contamined cultured blue mussels. Besides, from fields observations and translocation experiences to safe area, we have seeked for probable depuration kinetics of shellfishes having accumulated during summer 1986 high levels of DSP producing Dinophysis. Such preliminary studies may drive to further experiences giving more accuracy to the establishment of thresholds recquired by shellfishes depuration policy.

Cinétiques de décontamination en milieu contrôlé de moules toxiques (Diarrheic Shellfish Poison). Premiers résultats ArchiMer

Laboratory assays have been performed, using mussels contamined with Dinophysis, in order to assess potential depurations kinetics. Mussels samples from Seine bay, Douarnenez bay and Vilaine bay have been tested . From these assays, which are to be performed in 1989; two main results are stressed : 1) when the toxicity level is near 0,5 MU/g of hepatopancreas*, depuration is undedectable during twelve days, 2) when the toxicity level exceeds 1 MU/g,depuration is important during the first eight days. *(quarantine level used by Japanese authorities) .

Hydrologie et distribution de dinophysis en juin 1988, dans la frange côtière nord-Gascogne ArchiMer

During the oceanographic cruise of June 1988, phytoplankton and correlated hydrological parameters were studied in the coastal area between Camaret and Ile d'Oléron. 62 stations were sampled in the 0-30 m layer within the 50 m depth limited coastal area. Observations focused on two toxic dinoflagellate species: Dinophysis spp and Gyrodinium aureolum. Dinophysis spp is more abundant in the Loire/Vilaine area, which is.characterized by high surface Nitrate levels, high 30 m depth Phosphate levels and Chlorophyll a amounts higher than elsewhere during the survey. Le maximum saisonnier de Dynophysis cf acuminata à Antifer (Normandie, France) ArchiMer

Since 1987 Antifer oil tanker terminal (Normandy, France) was weIl known, as a preferential area for summer highest blooms of Dinophysis cf acuminata, a toxic dinoflagellate. Three oceanographic cruises were made in August 1989, before, in the course of and after the seasonal max. of this species. Results obtained from these cruises (hydrological parameters and phytoplankton) as weIl as from a daily survey at a reference station in Antifer harbour, corrobora te the prevailing role of coastal up flowing of Seine estuarine waters, pushed to the coast by a dominant S - SW wind, on intensity and duration of Dinophysis bloom. Such observations are in agreement with those of Summer 1987 at reference station.

Détection automatique par visionique des Dinophysis spp. contenus dans des échantillons d'eau de mer. Premiers résultats ArchiMer

Nous avons réalisé cette étude dans le but de fournir aux agents d'IFREMER chargés de la surveillance des eaux littorales et en particulier de la flore phytoplanctonnique, un outil permettant d'identifier et de compter les cellules de Dinophysis de façon automatique. La visionique nous semblant être une discipline qui se prêtait à ce type de détection, nous avons réalisé une étude de faisabilité d'un système d'analyse d'il}1age appliqué à la reconnaissance et au comptage de Dinophysis. Le complexe «Dinophysis acuminata»: identification des espèces le long des côtes françaises ArchiMer

From seawater samples collected between 1984 and 1989 in 12 French coastal areas, a morphometric study have been realised on Dinophysis genera restricted to "acuminata" like species. Simple morphological criterias are given as suitable for discrimination of four dominant species occurring during toxic episodes in shellfish farms (Diarrheic Shellfish Poisoning): Dinophysis c-closed-integral. sacculus, D. c-closed-integral. acuminata, Dinophysis sp. and D. c-closed-integral. norvegica. This last species cannot be included in the "acuminata complex" and produces temporary blooms along southern Brittany coasts. Pending a more precise diagnosis it is recommended to clarify the relationship between toxin composition and each species previously described.

Innovative rapid solid-phase immunobead assay for the detection of okadaic acid and related DSP toxins in shellfish ArchiMer

The solid-phase immunobead assay (Ciguatect super(TM)) has been optimized to detect toxins associated with diarrheic shellfish poisoning (DSP). The Ciguatect super(TM) test kit utilizes a membrane to bind the toxins and a monoclonal antibody-latex bead solution as the determination step. The presence or absence of okadaic acid (OA) and related toxins is determined by binding the toxins to a membrane attached to a plastic strip and exposing the toxins ladened membrane to a monoclonal antibody-colored latex bead complex which has a high specificity for OA and related polyether compounds. Application de modèles dynamiques bayésiens à la prévision des efflorescences à Dinophysis à Antifer (Baie de Seine) ArchiMer

L'objectif de ce travail était de mettre en place un outil mathématique permettant de prévoir les concentrations en Dinophysis cf. acuminata, micro-algue toxique. Nous avons décidé d'utiliser des méthodes dynamiques de prévision bayésienne : modèles polynomiaux du premier, second et troisième ordre, modèle polynomial du premier ordre avec intervention, et modèle de régression multilinéaire. Les modèles polynomiaux du premier, second et troisième ordre se sont avérés inadaptés. Le modèle de régression multilinéaire a été testé avec les variables concentrations en phosphate (PO4) et en nitrate (NO3). Dinophysis spp. cells concentrated from nature for experimental purposes, using size fractionation and reverse migration ArchiMer

A method based on size fractionation plus reverse phototactic migration was used to collect large Dinophysis spp.-dominated populations. Cells were active and survived handling and transportation. Le Dinophysis est une microalgue toxique à l’origine d’epidémies diarrhéiques. En dépit de nombreuses études, l’écologie et la biologie de ce genre phytoplanctonique restent mal connues. Afin de déterminer les conditions d’apparition de Dinophysis cf. acuminata sur le site d’Antifer (Normandie, France), des prélèvements d’eau de mer ont été effectués quasi-journellement pendant les périodes estivales des années 1987 à 1993. Les séries temporelles résultantes concernent la concentration de la microalgue et les facteurs chimiques, physiques et météorologiques. Classical regression analysis can be used to model time series. However, the assumption that model parameters are constant over time is not necessarily adapted to the data. In phytoplankton ecology, the relevance of time-varying parameter values has been shown using a dynamic linear regression model (DLRM). The marine dinoflagellate genus Dinophysis includes species that are the causative agents of diarrhetic shellfish poisoning (DSP). Recent findings indicate that some Dinophysis species are mixotrophic, i.e. capable of both autotrophic and heterotrophic nutrition. Ifremer monitoring network for phytoplankton and phycotoxins (REPHY) is in charge of shellfish safety control and carries out shellfish toxicity test on mice as soon as Dinophysis cells (okadaic acid "OA" producer) are observed in sea water. However, this test can neither identify or quantify the diarrhetic toxins OA and its analogues (DTX-1, DTX-3). Only a chemical analysis is able to detect them. First report on amnesic and diarrhetic toxins detection in French scallops during 2004-05 monitoring surveys ArchiMer

In the context of the French phytoplankton and phycotoxins monitoring network (REPHY), shellfish scallops were harvested systematically during the authorised fishing season for the purpose of investigating paralytic toxins (PSP-mouse test), amnesic toxins (ASPchemical analysis), and lipophilic toxins (DSP-mouse test).
